VAV1
VAV1,即Vav鳥嘌呤核苷酸交換因子1,別名VAV,是一種信號轉導蛋白,屬于Vav家族成員之一。VAV1主要在造血細胞中表達,包括T細胞、B細胞、單核細胞、自然殺傷(NK)細胞、粒細胞和樹突狀細胞。
VAV1的作用機制涉及作為Rho亞家族GTP酶的磷酸化依賴性的GDP/GTP交換因子(GEF),激活導致肌動蛋白細胞骨架重新排列和轉錄改變的信號通路。
VAV1在T細胞和B細胞的發育和激活中發揮重要作用,參與免疫細胞生長、增殖、轉化和遷移的調節。VAV1對T細胞受體激活的反應強度有重要影響,特別是鈣離子參與的激活轉錄因子等事件。
VAV1的異常表達與多種疾病相關,包括自身免疫性疾病和腫瘤惡化,使其成為治療和干預癌癥的潛在靶點。
