KLRC1
KLRC1,全稱為殺傷細胞凝集素樣受體C1,也被稱為NKG2A、CD159a、NK細胞受體A。KLRC1主要表達在NK細胞表面,屬于NKG2家族成員,與另一個家族成員KLRD1/CD94形成異二聚體,識別MHC I類HLA-E分子。KLRC1通過與HLA-E結合,負調控NK細胞、CD8+ T細胞等的免疫活性,參與腫瘤及病毒感染細胞的殺傷。研究發現,KLRC1在哮喘患者中表達上調,提示檢測KLRC1受體水平可以用于哮喘的臨床診斷、直接反映哮喘控制情況,以及KLRC1抗體運用于哮喘疾病防治的應用。目前,針對KLRC1的藥物研發主要集中在NKG2A抑制劑,例如莫那利珠單抗(Monalizumab)和阿斯利康開發的Interlink-1試驗。莫那利珠單抗是一種NKG2A抗體單抗,可阻斷NKG2A與HLA-E的相互作用,在多種癌癥適應癥中展現出良好的治療效果。然而,NKG2A抑制劑的上市還需要更大規模的臨床試驗進行驗證。
