SLC22A12
SLC22A12，即溶質(zhì)載體家族22成員12，別名尿酸鹽轉(zhuǎn)運(yùn)蛋白1（URAT1）、新型腎臟轉(zhuǎn)運(yùn)蛋白（NKT）。它是一種主要在腎臟近端小管上皮細(xì)胞的頂端刷緣膜上表達(dá)的跨膜蛋白，負(fù)責(zé)尿酸鹽的重吸收過程。SLC22A12的作用機(jī)制是通過利用管腔兩側(cè)的濃度梯度和化學(xué)梯度差，將尿酸鹽從近端腎小管腔內(nèi)轉(zhuǎn)運(yùn)到腎小管上皮細(xì)胞，這是尿酸鹽重吸收的第一步。SLC22A12在生物學(xué)上的意義主要體現(xiàn)在其對尿酸鹽平衡的調(diào)控作用。它參與調(diào)節(jié)血尿酸水平，影響尿酸的排泄和重吸收。SLC22A12的功能缺失突變會導(dǎo)致尿酸鹽重吸收功能障礙，可能引起1型腎低尿酸血癥。此外，SLC22A12中的單核苷酸多態(tài)性（SNP）與代謝綜合征的發(fā)病機(jī)制有關(guān)，包括肥胖、低高密度脂蛋白膽固醇、腰圍增加等。因此，SLC22A12是研究和治療與尿酸鹽代謝相關(guān)的疾病，如痛風(fēng)和高尿酸血癥的潛在靶點。
