L1CAM
L1CAM，全稱 neural cell adhesion molecule L1，是一種廣泛表達(dá)于神經(jīng)系統(tǒng)中的細(xì)胞粘附分子。它也被稱為CD171、L1、S10、MIC5等。L1CAM蛋白在神經(jīng)元遷移、軸突生長(zhǎng)、突觸形成等過(guò)程中發(fā)揮重要作用，參與構(gòu)建神經(jīng)網(wǎng)絡(luò)的精細(xì)結(jié)構(gòu)。研究表明，L1CAM蛋白與多種疾病的發(fā)生發(fā)展密切相關(guān)，包括癌癥、神經(jīng)退行性疾病等。因此，針對(duì)L1CAM蛋白的藥物研發(fā)成為近年來(lái)研究的熱點(diǎn)。目前，已有多種靶向L1CAM蛋白的藥物進(jìn)入臨床試驗(yàn)階段，顯示出在癌癥治療方面的潛力。

IHC image of CSB-RA588962A0HU diluted at 1:50 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.76% DAB.
Overlay Peak curve showing MCF-7 cells stained with CSB-RA588962A0HU (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1μg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1μg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
