GFRA1
GFRα1(膠質(zhì)細(xì)胞源性神經(jīng)營(yíng)養(yǎng)因子受體α1)是GDNF家族配體的核心受體之一,通過(guò)與RET酪氨酸激酶受體形成復(fù)合物調(diào)控神經(jīng)元存活與分化。該蛋白在胚胎發(fā)育期對(duì)交感神經(jīng)、腎臟及腸道神經(jīng)系統(tǒng)的形成起關(guān)鍵作用,其功能異常可能引發(fā)先天性巨結(jié)腸癥、多巴胺能神經(jīng)元退化等疾病。近年研究發(fā)現(xiàn)GFRα1在甲狀腺癌、神經(jīng)母細(xì)胞瘤中存在異常表達(dá),其介導(dǎo)的RET信號(hào)過(guò)度激活與腫瘤侵襲轉(zhuǎn)移密切相關(guān)。針對(duì)這一通路開(kāi)發(fā)的凡德他尼、卡博替尼等RET抑制劑已獲批用于甲狀腺髓樣癌治療,而基于GDNF-GFRα1系統(tǒng)的基因療法在帕金森病動(dòng)物模型中展現(xiàn)出神經(jīng)保護(hù)潛力。目前重組GDNF蛋白的鞘內(nèi)給藥方案正在進(jìn)行Ⅱ期臨床試驗(yàn),旨在為肌萎縮側(cè)索硬化等神經(jīng)退行性疾病提供新治療策略。
